cOFM – cerebral OFM 

Sampling Beyond the Blood Brain Barrier

  • Membrane free, push-pull probes with macroscopic openings are used to sample interstitial fluid (ISF) within cerebral tissue to provide a dynamic in vivo metabolic profile of the brain. 

  • This enables the analysis of peptides, proteins, antibodies, lipophilic substances, bound and unbound drugs, enzymes, nanocarriers, vesicles, cells, and any other molecules regardlress of molecular size or lipophilicity. 
  • The blood brain barrier (BBB) is re-established within 14 days after implantation via a healing dummy insert. Once the BBB is healed and the healing dummy insert is replaced with a sampling insert, cOFM allows for sampling in brain tissue with an intact and healthy BBB, continuous monitoring of drug transport accross the intact BBB, and dosing beyond the BBB. 
  • ISF sampling is possible in concisous or anesthetized animals, with continuous sampling possible up to 30 days.

Microperfusion Pump

This Microperfusion-Pump is a push-pull pump designed specifically for use with the OFM probes and accessories for ISF collection during preclinical and laboratory studies.

Concentric OFM Guides, Healing Dummies, and Sampling Inserts

Procedures require one OFM Guide Probe Kit, and one corresponding Sampling Insert.

Each Concentric cOFM Guide Kit includes: an OFM guide (1), a corresponding healing dummy insert (2), and a locking wedge (4). Extra healing dummy inserts (see product chart below) and locking wedges can also be purchased separately. Sampling inserts (3) are sold individually.

The guide should be implanted with the healing dummy insert, and remain in place for 14 days to allow the BBB to recover. Once the BBB is healed, animals should be sedated, and the healing dummy replaced with a sampling insert. Instructions for Use cOFM Probe Kit

Selecting Guide and Insert Sizes:

When selecting the cOFM guide size, the ventral stereotaxic coordinates need to be calculated so that the open surface length of the guide (area Y in the diagram below) is inside or immediately adjacent to the region of interest after implantation. NOTE: For those familiar with using a Microdialysis guide cannula, this is the change. No extra space is required for the sampling insert, and therefore, the ventral stereotaxic coordinates should be re-calculated to ensure the open surface of the OFM guide itself is directly within the sampling area.

Guides are available with either 1mm or 2mm open surface lengths. The sampling insert (and any replacement healing dummy inserts) need to correspond to the total length of the guide, including the open surface length and the shaft length (Y + X in the diagram below).

Use the diagram and chart below to select your OFM Guide Kit and associated sampling insert (and extra healing dummy insert if needed).

  • Inserts ranging from 2-8 mm are ideal for mouse and juvenile rodent dorsal and ventral region sampling.
  • Inserts between 7-10 mm are ideal for rat dorsal and ventral region sampling.
  • Inserts up to 22 mm are ideal for sampling in pigs and other large animals.
  • Replacement healing dummy inserts and locking wedges can be ordered if needed. 
  • Custom insert sizes can also be requested.

Tubing

Low-bind tubing has been designed specifically for open flow microperfusion (OFM) studies. Compared to traditional FEP or PEEK tubing, this fluropolymer tubing is less gas permeable and minimizes air bubbles optimizing flow and allowing for increased recovery rates.
